I'm excited about Trisquel 3.0.

I'd like to use it in my classes for audio and video production. Is there a way I can get it to work with a real-time kernel?

By now, the rt kernel is available in the 2.x branch only. But if you
are going to use it for teaching, then I'd recommend you to use Trisquel
2.2 Edu, it has several improvements for schools. To get a realtime
kernel on it, just install the linux-image-rt package.

Anyway, in my experience a fully preemptible realtime kernel is not
needed, as the latency is mostly the same as with the generic kernel
-which is still voluntarily preemptible-, and the rt one is less stable.

I'm no expert in rt though, just an amateur audio enthusiast. :)
